The New Zealand actor, born in Northern Ireland, was best known for his leading roles in the dinosaur blockbusters and "The Piano", and appeared in over 50 films during his career.
The family of the film star, who was one of New Zealand's most successful in Hollywood, said on Monday that his death was "sudden and unexpected".
In 2022, Neill was diagnosed with stage three angioimmunoblastic T-cell lymphoma, a rare type of non-Hodgkin lymphoma.
After the cancer diagnosis, Neill said he would take a short break from acting, and earlier this year announced that he was healthy.
His family reported that he died on Monday in Sydney, Australia, but did not specify the cause of death.
"Sam was surrounded by family and passed with the dignity that characterized his entire life," they said.
