Messi, who helped "Miami" win the 2023 title in the first weeks of his time with the Florida club, increased his scoring tally in this tournament for clubs from Major League Soccer (MLS) and Mexico's Liga MX to a record 14 goals.
David Rodríguez put "San Luis" ahead in the fourth minute, but Messi equalized seven minutes later.
Venezuelan midfielder Telasco Segovia put "Miami" ahead in the 26th minute.
Messi scored again shortly before halftime, when Brazilian defender Micael made it 4–1 in stoppage time at the end of the first half.
Atlético's Rafa Llorente scored a consolation goal in the second half.
